I have not used the Photo Studio 6 software yet, so cannot express any opinion on it. I have a bit more testing to do before I start our scanning and storage project of 30+ years of pictures/ negatives (many 1000's), but am happy with the quality of the scans and the versatility of various settings within the respective modes. The actual transparency adapter in the v500 is large. The only difference between the two scanners in this regard is that the v600 comes with a different 120 film holder, that has a larger opening that the 120 holder that comes with the v500. I have experimented enough and read enough other user comments to feel that the Home setting will likely be my go-to mode as the Auto mode does not seem to auto-recognise the separation between the different photos when( scanning 2 or 3 at a time and did not seem to scan the negatives at all and kept producing an image of the film holder only. The V600 is advertised as being able to scan more 120 negatives, but this really isn't entirely true.
